Determination 2 - Isatuximab, Iberdomide, Bortezomib and Dexamethasone Induction, Followed by Risk- and Response-Adapted Consolidation and Maintenance Therapy, in Transplant-Eligible Patients With Newly Diagnosed Multiple Myeloma

What's the purpose of the trial?

The purpose of the study is to determine the best treatment approach based on the risk profile of the cancer cells and on how the disease responds to treatment. This is a randomized research study evaluating treatment for transplant-eligible participants with newly diagnosed multiple myeloma. Induction therapy in this study includes the drugs isatuximab, iberdomide, bortezomib, and dexamethasone. After induction therapy, participants will receive consolidation and maintenance therapy that is adapted based on their risk profile and response to treatment. The research study procedures include: screening for eligibility, study visits, blood and bone marrow tests, disease assessments, treatment with study drugs, and follow-up visits. It is expected that about 720 participants will take part in this study.

Experimental Treatments

  • Isatuximab is a type of drug called a monoclonal antibody. Isatuximab targets a protein called CD38, which is present in high numbers on the surface of multiple myeloma cells, as well as on certain other types of cells, such as red blood cells.
  • Iberdomide Investigational drug Iberdomide (CC-220) is a Cereblon (CRBN) binding compound currently under clinical investigation for multiple myeloma.
  • Melphalan is a chemotherapy medication used in the treatment of different cancers.
  • Linvoseltamab is a bispecific antibody being tested for use in multiple myeloma that targets BCMA on cancer cells and CD3 on T-cells . 
  • Bortezomib Bortezomib is a kind of medication called a proteasome inhibitor that interferes with the growth and spread of cancer cells in the body. Bortezomib is used to treat multiple myeloma and mantle cell lymphoma.
  • Dexamethasone is a corticosteroid that prevents the release of substances in the body that cause inflammation, and is given in conjunction with some cancer treatments.
